  • PRIM-SP: Privileged structures - Natural-like compounds from keto-acids.(2008)

    Mission :

    The project aims at designing and synthesizing chemical libraries to enrich the regional library with privileged structures inspired from natural products.

    Objectives and Rationale:

    Our regional screening library is a set of 45000 either lead-like, drug-like molecules or fragments (for structure-based discovery). In order to improve the set of more sophisticated compounds, that display for example quaternary centers and controlled chiral carbons, we develop the synthesis of privileged structures from a common template: keto-acids. These precursors allow the synthesis of enantiomers or diastereoisomers either in racemic or pure chiral form of piperidines, piperazines, lactams …A strong emphasis is put on green technologies using solvent-free microwave procedures or water based protocols. The main reactions used are the Ugi multicomponent reaction and the Meyers’ reaction a binucleophile-bielectrophile reaction type.
